Photodynamic therapy (PDT) is a clinically approved non-surgical therapeutic alternative involving the use of a photosensitizer activated by an appropriate light source for the treatment of neoplastic and non-neoplastic diseases. In this study, a proteomic approach was used to characterize protein expression profiling in human keratinocytes after applying PDT using chlorin e6 (Ce6) as a photosensitizer. 13 differentially expressed proteins showed changes in expression level of at least 1.5-fold by Ce6-mediated PDT. Of these, 12 protein spots were down-expressed, while 1 protein spot was up-expressed by PDT. The proteomic result for expression of pyruvate kinase M2, elongation factor Tu and ezrin was consistent with that obtained by western blot. Especially, the expression of glycolytic pyruvate kinase M2 was down-regulated, and the lactate level produced was reduced by Ce6-mediated PDT. These proteins might be useful to develop photo-cytotoxic biomarkers and also provide insight into biological signals underlying the systematic responses of cells to Ce6-mediated PDT.
